Resident Evil: Caliban Cove is the second novel of S.D. Perry's novelization series. Set between the events of Resident Evil and Resident Evil 2. Due to the establishment of Resident Evil 2's presence within the timeline being in 1999's Resident Evil 3: Nemesis, the Caliban Cove timeline is inconsistent with its video game counterpart's.

Set in the aftermath of their ordeal through the Umbrella Corporation's genetic research facility, the surviving members of the S.T.A.R.S. attempt to warn the world about the conspiracy to create terrifying biological weapons. But the conspiracy is far from dead, as they learn when they are declared outlaws by the very people who trained them. Forced to go underground, the S.T.A.R.S. resolve to battle the conspiracy on their own, determined to seek out and stop Umbrella's experiments wherever they may be.

Combat medic and biochemist Rebecca Chambers, the sole survivor of Bravo Team, joins a new S.T.A.R.S. strike force when rumor comes of another Umbrella experiment: hidden beneath the rocky cliffs of Caliban Cove, Maine, someone is building an army of the undead. Plot

In the days following the destruction of the Spencer Mansion, Raccoon City's S.T.A.R.S. team has been disbanded for their "blatantly unprofessional conduct", the deaths of their comrades blamed on a drug-related helicopter crash. The apparent stop in further killings has been linked to the mansion's explosion, an explanation given by Chief Irons as the gang starting a campfire in the abandoned building and causing a chemical explosion.

In the aftermath, Barry Burton has taken charge of the now-unofficial Raccoon branch. S.T.A.R.S. HQ in New York has been briefed on the prior events and has promised to lead its own investigation, but they have remained quiet for nearly two weeks. With new developments in the investigation via his own channels, Barry orders Chris; Jill and Rebecca to meet him at his house, Brad having left town two days earlier. Arriving first, Rebecca is introduced to David Trapp, Captain of the S.T.A.R.S. Exeter branch. He informs her of the situation: Umbrella has successfully bribed a number of high-ranking S.T.A.R.S. leaders to end the investigation, and he fears his own Exeter team has been turned. Beyond Assistant Director Kurtz being under Umbrella's direct control, S.T.A.R.S.' figure head and founder, Dr. Marco Palmieri, is being rushed for early retirement to remove his influence on the remaining S.T.A.R.S. teams. On top of the collapse of S.T.A.R.S.' investigation, David announces his discovery of an Umbrella base in Maine which has also experienced a biohazard.

David enlists the Raccoon S.T.A.R.S. on a mission to infiltrate the coastal Caliban Cove facility, obtain proof of Umbrella's actions and leave. The unit is still small, consisting of the four Raccoon members; David, and the three people in Exeter he can trust. During preparation for the mission, Barry begins making calls to friends in S.T.A.R.S., hoping to find more allies; Chris meanwhile gets in touch with the FBI in hopes of an internal investigation into Chief Irons' conduct in the RPD. David continues, revealing his participation in the investigation was due to the meddling of Trent, a mysterious man who Jill had met before and who was willing to provide information on Umbrella. David shows the team a list of Umbrella researcher names he was provided with. William Birkin is one name Jill knows from the same list as Trent gave her before the mansion. One of the names Rebecca knows as Dr. Nicholas Griffith, a former biochemist that had disappeared after being fired for unethical experimentation.

The house is attacked by unknown assailants, forcing them to all run out into the woods to hide. David kills one of them on the way out, recognising him as Jay Shannon, Captain of the Oklahoma City branch. With the RPD's K-9 unit heard barking in the distance, the group take refuge in Brad's empty home. With Umbrella knowing him to be away, they wouldn't check there. After a little talk, Rebecca and David set out and leave on a private airplane for Exeter.

At Caliban Cove Dr. Griffith stands atop a lighthouse, musing on his latest and greatest accomplishment; a virus that robs a human of their ability to think, they become servants to whomever orders them. Griffith has already used his new virus on three other scientists at the Cove facility, and none of them can make any decisions outside of what Griffith tells them. He muses that once he gives his "gift" to the wind, everyone will be free.

At Exeter, the home of S.T.A.R.S. forensics officer Karen Driver, David briefs his own team; comprised of himself, Rebecca, Steve Lopez, John Andrews, and Karen. They set out, collecting the equipment they need, and begin the Caliban Cove mission, with no back up.

Moving towards Caliban Cove using a raft, the team comes under attack by an unknown creature that destroys the raft, stranding the team in the cove. Moving into a dock house, they find a body nailed to the door opposite, it's hand in a waving position. After searching around, they discover the body is that of Dr. Kyle Ammon, a name in the list of people Trent gave them. They soon come under attack by Tri-Squads, zombies that have use of basic intelligence and M-16s. The team escapes after defeating one of the four squads.

Throughout the course of the mission, Karen gets infected by the T-Virus and succumbs to it, while Griffith and another researcher, Alan Kinneson, manage to capture Steve and inject him with a virus. David, Rebecca, and John eventually get captured by Steve, who is ordered by Griffith to execute himself. Recovering a "lucky charm" pineapple grenade from Karen's corpse, David, Rebecca, and John manage to free themselves from captivity and escape the facility while Griffith dies from the explosion.

The survivors of the Exeter team are recovered by a Captain Blake from the Philadelphia S.T.A.R.S. branch, who were deployed at the request of Barry.
